ABSTRACT

Inwardly rectifying potassium (Kir) channels exhibit an asymmetrical conductance at hyperpolarization (high conductance) compared to depolarization (low conductance). The KCNJ10 gene which encodes an inwardly rectifying K+ channel Kir4.1 subunit plays an essential role in the inner ear and hearing. Mutations or deficiency of KCNJ10 can cause hearing loss with epilepsy, ataxia, sensorineural deafness, and renal tubulopathy (EAST) or SeSAME (seizures, sensorineural deafness, ataxia, mental retardation, and electrolyte imbalance) syndromes. In this review, we mainly focus on the expression and function of Kir4.1 channels in the inner ear and mutation-induced EAST/SeSAME syndromes to provide insight for understanding the pathogenesis of deafness induced by KCNJ10 deficiency.

ABSTRACT

OBJECTIVE@#To analyze the MR imaging findins of acinic cell carcinoma(ACC) in parotid gland and develop the diagnosis of this disease.@*METHOD@#The MR imaging fearures of 11 patients with pathologically proved ACC were retrospectively analyzed. MR imagings were analyzed in relation to the follow:amount, location, size, shape, margin, pseudocapsule, MR signal intensity and enhanced pattern and the cervical lymphadenopathy. The clinical analysis on patients included age, sex, and follow-up.@*RESULT@#There were 11 patients, 7 lesions was located in the right parotid gland and 4 lesions was located in the left parotid gland; the average maximum diameter was (2.66±0.99)cm; 7 lesions showed lobulated,and 4 lesions showed round; 8 lesions had no pseudocapsules and 3 lesions had incomplete pseudocapsule. All lesions showed homogeneous or heterogeneous isointense and slight hyperintense on T1 WI and T2WI. On post contrast images, the tumor parenchyma ingredients showed remarkable enhancement in all lesions.@*CONCLUSION@#The MR imaging of ACC in the parotid gland don't have specific features, but when the tumor of the parotid gland showed no pseudocapsule (or showed incomplete pseudocapsule), lobulate, small cysitc and remarkable enhancement, it may indicate ACC, and the correct diagnosis depends on clinical pathology.

ABSTRACT

Objective To evaluate the rat model of type 2 diabetes mellitus ( T2DM) associated with depres-sion by body weight, fasting blood glucose, manifestations, and open field test.Methods The T2DM rat model was induced by high fat diet and low dose of STZ injection, and in addition, the T2DM rats were made restraint stress for 21 days.32 Wistar rats were randomly divided into three groups: normal group ( group N) , T2DM group ( group T)and T2 DM with depression group ( group T +D) , with 8 rats in each group.After the model was established, to measure the body weight, fasting blood glucose at day 0, 7, 14, and 21, and observe the gross manifestations, drink-ing and diet, feces, urine, and mental state, and test the rat depression by open field test.Results After establish-ment of the T+D rat model, the rats in group T+D showed some symptoms, including messy dark and gloomy hair, slow movement, increasing drinking, diet, feces and urine and mental fatigue.At day 0, 7, 14, and 21, compared with the group N, the body weight of the group T and group T+D were decreased, showing a significant difference ( P0.05).Compared with the group N, the movement speed in 5 minutes in the group T was significantly slower than that in the group T , with a significant difference ( 5.1 ±0.4 cm/s vs.2.9 ±0.6 cm/s, P<0.05 ) , and even more slower than that in the group T +D, with a significant differ-ence (5.1 ±0.4 cm/s vs.2.4 ±0.5 cm/s, P<0.01) .Conclusions A rat model of type 2 diabetes mellitus as-sociated with depression has been successfully established by high fat diet and injection of low dose streptozotocin in combination with restraint stress for 21 days.This rat model is useful for further relevant studies.

ABSTRACT

ObjectivesTo identified the strain 1012 from the National Center of Clinical Laboratory of China for microbe inter-laboratory quality assessment in 2010, and study the taxonomic status of strain 1012 and related species in the genus Actinomyces. Methods The bacterial traditional morphological characteristics, commercial API systems, and 16S rRNA gene sequence analysis were applied to identify the problematic culture of strain 1012. The phylogenetic tree based on the remote information of the prokaryotes systems was constructed to study the taxonomic status and evolutionary relationship of the genus Actinomyces and related species in the family Actinomycetaceae. Results Strain 1012 was determined as a kind of facuhative anaerobic,non-spot-forming,Gram-positive coryneformbacteria,which was identifiedto Actinomyces turicensis for the phenotypic biochemical characteristics of more than 60 items, The comparative study of 16S rRNA gene showed the strain 1012 with 99. 8% similarities to Actinomyces turicensis, but only 90. 6% to the type species of Actinomyces bovis in the genus Actinomyces. However, the comparative study of 16S rRNA gene showed the strain 1012 with only 90. 6% homology to the type species of Actinomyces bovis in the genus Actinomyces. Further phylogenetic analysis showed that nine independent clusters were grouped in the family Actinomycetaceae, of which four clusters were separately represented the genera Varibaculum,Mobiluncus, Actinobaculum and Arcanobacterium, while other five clusters all were designated to the genus Actinomyces. The study showed strain 1012 was located in genus Ⅲ of Actinomyces, yet with a relatively long genetic distance to Actinomyces bovis. ConclusionThe genus Actinomyces may be reclassified as one genus Actinomyces sensu stricto and several new genera for the genotypic characteristics.

ABSTRACT

Objective To observe the clinical efficacy of orthodontic treatment for displacement of anterior tooth caused by periodontal disease. Methods Forty-seven patients with displacement of anterior tooth caused by periodontal disease were divided into study group (26 cases) and control group (21 cases)according to the therapy method, the study group received periodontal basic treatment combined with orthodontic treatment, while the control group only received periodontal basic treatment, the changes of periodontal pocket depth, bleeding index, anterior coverage, overbite, alveolar bone height in two groups before and after treatment were observed and compared. Results The periodontal pocket depth, bleeding index, anterior coverage and overbite of study group after treatment were (3.12±0.53) mm, (27.30±4.25)%, (2.53 ± 0.76) mm and (2.18 ± 0.53 ) mm respectively and which were significantly lower than those before treatment [(5.35 ± 0.68 ) mm, (97.60±7.53 )%, (6.22±1.43 ) mm, (4.53±2.25)mm], the difference was statistically significant (P< 0.05 ), and the decrease extent was more obvious than the control group, the difference between two groups was statistically significant(P<0.05); the above indicators in control group after treatment also dropped, but compared with those before treatment there was no significant difference (P >0.05); the alveolar bone height in both groups had no obvious change before and after treatment.Conclusion Periodontal basic treatment combined with orthodontic treatment can correct the malposed anterior teeth, improve the periodontal conditions, it has good application prospects.

ABSTRACT

@#Objective To investigate the effect of both conservative and surgical treatment on cervical spinal cord injury without fracture and dislocation.Methods28 patients were divided into the surgical group (16 cases, treated with modified open door laminoplasty using anchor method and carried on early rehabilitation training) and conservative group (12 cases, treated with rehabilitation training after admission). All patients were assessed with JOA grade before and 3 months, 6 months and 12 months after treatment, and improvement rate was calculated.ResultsJOA grade of all patients increased after both surgical and conservative treatment ( P< 0.001), but the effect of surgical treatment was significant better than that of conservative treatment ( P< 0.001).ConclusionThe effect of early surgical treatment on cervical spinal cord injury without fracture and dislocation is superior to conservative treatment.

ABSTRACT

Objective To investigate the application of three dimensional blood vessel angiography(3D-CTA) in the patients of spontaneous subarachnoid hemorrhge diagonised as aneurysm.Methods 3D-CTA were examined in 32 cases of spontaneous subarachnoid hemorrhage.Results In 32 patients of spontaneous subarachnoid thirty patients were diagnosed as aneurysm.Two cases were not diagnosed.Conclusion 3D-CTA may be the first examination method in spontaneous subarchnoid hemorrhage.

Objective To detect the expression of AKT2 in human colon cancer and understand its relationship with PTEN.Methods The expressions of AKT2 and PTEN were detected in 30 patients with normal colonic tissues,30 patients with colon adenoma tissues,and 64 patients with colon carcinoma tissues by immunohistochemical SP staining method.Results The positive expressive rate of PTEN presented a trend of progressive decrease from normal tissues,adenoma tissues to colon carcinoma tissues,while the positive expression rate of AKT2 presented a trend of progressive increase.PTEN expression was obviously higher in normal colon tissues than in colon adenoma tissues and colon cancer tissues(?2=68.855,P

ABSTRACT

Objective The prestin,a motor protein responsible for outer hair cell(OHC)electromotility,is expressed on the OHC surface.Previous experiments revealed that OHC electromotility and its associated nonlinear capacitance was mainly located at the OHC lateral wall and was absent at the apical cuticular plate and the basal nucleus pole.Immunofluorescent staining for prestin failed to demonstrate the prestin expression at the OHC basal ends in whole-mount preparation of the organ of Corti.The aim of this study is to investigate the distribution of prestin at OHC.Methods In this experiment,the localization of prestin protein in single dissociated OHCs from cochlea of normal mouse,rat and guinea pig,were examined by immunofluorescent staining and confocal microscopy.Results We found that prestin was uniformly expressed on the OHC basolateral surface,including its basal pole.No staining was observed on the cuticular plate and stereocilia.The OHC lateral wall had a trilaminate organization and was composed of the plasma membrane,cortical lattice,and subsurface cisternae.By with co-staining with a membrane marker di-8-ANEPPS,prestin-labeling was found locating at the outer layer of the OHC lateral wall.Further separating the plasma membrane from the underlying subsurface cisternae,using a hypotonic extracellular solution,prestin-labeling was shown locating at the plasma membrane instead of the subsurface cisternae.Conclusion The data revealed that prestin is expressed in the plasma membrane on the whole OHC basolateral surface.

ABSTRACT

Objective:To study the effect of 5-FU polylactic acid nanospheres(5-FU-PAN)on the proliferation and apoptosis of the human tongue squamous cell carcinoma Tca83 cells.Methods:Tca83 cells were exposed to 5-FU-PAN or 5-FU at the doses(mmol/L)of 1?10-2,2?10-2 and 4?10-2 for 1,3,5,7,9 and 11 days respectively.The survival rates of the cells were examined by MTT assay,the apoptosis of the cells was observed by acridine orange fluorescent staining and the cell cycle distribution was studied by flow cytometry.Results:Both 5-FU-PAN and 5-FU inhibited the proliferation and promoted the apoptosis of the cells dose and time dependantly.5-FU-PAN showed stronger effects than 5-FU.The two agents blocked the cells at S phase and 5-FU-PAN was more potential than 5-FU.Conclusions:5-FU-PAN is more effective in the growth inhibition and apoptosis promotion of Tca83 cells.
